Growth of two blue-stain fungi associated with Tetropium beetles in the presence of callus cultures of Picea abies

Abstract

The callus tissue can be used to evaluate the potential ability of microorganisms to cause disease.The blue-stain fungi,Grosmannia piceiperdaandOphiostoma tetropiiare important associates ofTetropiumspp. inPoland. The opinions about their virulence are controversial. Here, we examined the growth mycelium of theG. piceiperdaandO. tetropiiin presence of the non-embryogenic cultures of Norway spruce, and accumulationof soluble and unsoluble proteins in this callus.The growth mycelium of one isolate ofG. piceiperdawas significantly stimulated whilst another isolate of thefungus and both isolates ofO. tetropiiwere unaffected by the presence of the callus. The significant higher(P<0.05) amount of soluble protein, was noted in the callus with both isolates ofG. piceiperda. In contrast toG. piceiperda, the callus withO. tetropiihad a similar concentration of soluble protein as the control. The im-portance of these results with respect to the pathogenic abilities and the in vivo behaviour of the examinedfungi is discussed
