Przydatność różnych metod in vitro w rozmnażaniu szparagów ozdobnych
AbstractNew species and cultivars of asparagus, recently introduced into the market, increased the diversity of the so-called cut greenery. However, their propagation by seed does not provide material of sufficiently standardised quality. To meet that requirement, propagation by tissue culture is increasingly used. Experiments were carried out on three cultivars of Asparagus densiflorus: `Sprengeri', ‘111leyeri' and `MyrioclarluS', on A. setaceus 'P3rramidalis' and on A. asparagoides 'Myrtifolius'. For the investigated cultivars the conditions of micropropagation by different methods were established. The best initial explants and the compositions and kinds of media (liquid or solid) were determined. A. densiflorus `Sprengerf was propagated using wdllary bud method, via direct and indirect (through callus stage) organogenesis and by somatic embryogertesis. Somatic embryos were obtained from callus initiated on the dicamba-containing medium, and then matured on the media with ancymidol or ABA, aminoacids, NAA, kinetin, or zeatin. All these stages needed to be done on Phytagel-solidified media so that the embryos could convert properly. On average, 73-469 embryos from 1 g of callus were obtained, and 1-70 % of embryos germinated properly. In A. densiflorus `Meyeri' adventitious shoots regenerated easily, but rhizogenesis did not occur. The 'Myriocladusr cultivar regenerated the best, especially by indirect organogenesis. The other investigated cultivars formed shoots with a high efficiency, but showed poorer results with roots (only 1-10 Lit, of rooted shoots), The in vitro-regenerated asparagus plants did not differ in phenotype and genotype from the traditionally propagated control plants.
