Niektóre właściwości glukoamylazy i innych glukozydaz towarzyszących jej w grzybowych preparatach glukogennych
AbstractCertain properties of gluociamylase,or -glucosidase /tranaglucosylase/ and α -amylase present in enzymic preparations from Aspergillus awamori, Endomycopsis capeularis and Rhizopus nigricans were investigated. The study was proceeded by an examination of some new improved methods of glucoamylase and transglucosylase activities determination. The preparations were subjected to some analyses and the glucosidases were separated by means of gel filtration on sephadex and DEAF-cellulose chromatography. It was found, that in Aspergillus awamori preparation two isoenzyace of glucoamylase, two forma of α -glucosidase /transglucosylase/ and one form of -amylase were present. The glucoamylase of this mould is an acid-stable enzyme, While the two other enzymes are destroyed at pH range 2,3-3,0. KI value for that glucoamylase with the soluble starch substrate equals 10,5 x 102 g/l, optimum pH 4,6, optimum pH = 4,6 optimum temperature being 65°C. In the Endomycopsis capsularis preparation two forms of glucoamylase, one form of α -glucosidase /transglucosylese/ and minute quantities of α -amylase were found. In this preparation the glucoamylase was also a more acid-stable enzyme than two other enzymes, which were inactivated at pH 3,0. Km value estimated for the glucoamylase with the starch substrate was 3,0 x 10-2 g/l, optimum pH was 5,6, optimum temperature was 55°C. It was Shown, that Rhizopus nigricans preparation did not contain say otherot -glucosidases except glucosmylase. The optimum temperature for this enzyme activity was 550C, optimum pH ranged between 4,1- 4,6 and Km value estimated with the starch substrate was 6,0 x 10-2 all . The difficulties in activity estimation of α-glucosidase and transglucosylase werediscussedvend a suggestion was made for an improvement of the procedure. In any case the action of tranaglucosylase was always accompanied by that of α-glucosidase. There is no doubt, that both the reactions - in the examine- tad preparations - are catalyzed by the same enzymic protein.
